Study Links Hypothalamic Menin Decline to Aging and Cognitive Impairment in Mice

Researchers from Xiamen University reported new findings suggesting that a protein called Menin may play an important role in regulating aging processes in mice.The study, published in PLOS Biology, focused on the hypothalamus, a brain region involved in metabolism, hormones, sleep, and stress regulation.Scientists observed that Menin levels declined significantly in aging mice, particularly in neurons located in the ventromedial hypothalamus.

This reduction was associated with increased neuroinflammation, memory impairment, reduced bone density, thinning skin, balance problems, and shorter lifespan.

To better understand the effect of Menin loss, researchers genetically reduced the protein in younger mice and found that they developed several premature aging characteristics.

The team also discovered that declining Menin levels reduced the production of D-serine, an amino acid and neurotransmitter linked to learning and memory.D-serine synthesis depends on enzymes that appear to be regulated by Menin.In another experiment, scientists restored Menin expression in older mice by delivering the gene directly into the hypothalamus.After approximately one month, the animals showed measurable improvements in memory, balance, skin thickness, and bone density.Supplementation with D-serine alone improved cognitive performance but did not reverse physical signs of aging.

Although the findings are considered promising, the research was conducted exclusively in mice, and scientists caution that it is still unclear whether similar treatments would be safe or effective in humans.The study contributes to growing evidence that the hypothalamus may play a central role in controlling biological aging.
